Methodology

Quantitative Reverse Transcription Polymerase Chain Reaction performed on the Cepheid GeneXpert instrument. This assay accepts Bone Marrow (BM) aspirate and Peripheral Blood (PB) specimens. This testing method can detect and quantify NPM1 mutation types A, B, and D only. All other mutation types will generate a falsely low signal or a false negative result.

 

The sensitivity of this NPM1 assay is 1:50,000 cells (NPM1-positive AML cells:control cells) or 0.03% (NPM1/ABL1 transcript percentage).

Specimen Requirements

Whole blood: Two (2) full 6mL EDTA (pink or purple) tubes required

Bone marrow: 3mL collected in EDTA (pink or purple) tube

 

Specimens must be received by lab as soon as possible, preferably within 24 hours of collection.
